Why a second set at all?
Next to che and cui, which never change, there is il quale, la quale, i quali, le quali. The meaning is the same. The difference: the ending shows the gender and number of the word it refers back to, and the sentence sounds more formal. You meet il quale mostly in writing, in the news and in speeches, rarely in conversation.
English has a formality switch of its own: the person I spoke with is everyday, the person with whom I spoke is careful and written. il quale sits roughly where whom sits. But in Italian the preposition is in front either way, in con cui and in con la quale. Only the choice of pronoun carries the register.

The four forms
il quale is built from an article plus quale. Both parts agree with the head noun, the word in front of the relative clause, and not with the role the pronoun plays inside the clause. English who and which give you no habit to lean on here.

After a preposition
il quale is most common after a preposition, as a more formal alternative to cui. The preposition merges with the article: di + il quale becomes del quale, a + la quale becomes alla quale.

When the reference would be unclear
When two nouns could be the head of the relative clause, che cannot say which one is meant. il quale can, through gender and number. This usually happens after di, con or per. English has no such tool: Marco's sister, who lives in Milan stays ambiguous, and you have to rewrite the sentence.
